Experimental Study of Furosemide on the Stria Vascularis, Organ of Corti and the Spiral Ganglion in- the Guinea Pig.





Abstract
Furosemide has been widely used as a loop diuretics in the field of clinic. Recently, Mathog reported furosemide has the toxic effects such as hearing loss and tinnitus. Brown studied on the pathophysiology of these side effects by means of electro-physiological approach.
However, the exact site and morphological change of the toxic effects on the hair cells of the organ of Corti are still obscure. In the present study, an experimental study was performed to observe the patterns and sites on the hair cells caused by.. administration of furosemide. Ten healthy guinea pigs weighing from 2009 to 3009 were used. Following administration of furosemide, the guinea pigs were sacrificed and fixed with 1% osmic acid. Morphological finding of the hair cells were observed under the surface preparation methods of cochlea.
The nuclei of outer hair cells were more markedly changed than the inner hair cells. The significant changes in the receptor organ of Corti were mainly noted near the round window of the inferior portion of the 2nd turn. The fusion of the stereocilia of hair cells were observed together with the partial loss of them. Also, many vesicles and lysosomes were observed in the subcuticular area and near the nucleus. The superficial layer ;of stria vascularis faced with endolymph showed the swelling of serectory cells. The significant changes in the outer hair cells of the organ of Corti were mainly noted near the oval window of basal turn and the inferior portion of the first turn of the cochlea.
